The 'X Factor' star will present the ITV1 show, which will explore quirky, funny and recognisable disputes between real-life couples.
The seven-part series - which will include a panel of comedians who all have their say on who should win each argument and "offer their own wit, wisdom and unique opinions" - was originally created by comedian Jerry Seinfeld and launched in America last year and ITV can't wait to remake it.
ITV1's commissioning editor for comedy and entertainment Claire Zolkwer said: "'The Marriage Ref' takes a close look at every day disagreements faced by couples - something we can all relate to and laugh with.
"It's an entertaining format and Dermot O'Leary will make a great host, not only giving his opinion but also leading the debate amongst the panel."
Annabel Jones, managing director at Zeppotron, which is producing the show, added: "This show promises to be filled with comedy moments - marriage and relationships can be unpredictable and hilarious. Dermot is a fantastic host with wide appeal and we'll be bringing some of the biggest names in comedy and entertainment to the panel. We're extremely proud to be making a series that was originally conceived by the comedy legend Jerry Seinfeld."
Dermot had previously spoken of his desire to host the show, saying: "It's a great show and the kind of thing I want to present because it's not your usual show. It's something I'd watch. I'd never do a gameshow. ITV have big plans for it, so fingers crossed."
