The White Stripes frontman - who was set to perform new James Bond theme 'Another Way To Die' with Alicia Keys at the event - is still recovering from an injury he suffered on his US tour last month.
A source said: "Jack is still in a lot of pain. He hoped it would have cleared by now but the injury is refusing to heal.
"MTV are disappointed as are Bond film chiefs. No one knows when he'll be fit enough to travel again."
The line-up for the Liverpool event - to be held in the UK city on November 6 - includes Beyonce Knowles, The Killers and Take That.
Alicia recently revealed she is keen to collaborate with Jack again because she was so pleased with their efforts for the 'Quantum of Solace' theme song.
She said: "This isn't the last time we'll work together, this is just the beginning.
"It's the first duet in Bond history and that's what makes it so fly - two worlds coming together to create something that totally goes with the movie."
